Carotid Intima Preparation

Carotid intima preparation is a tissue-dissection technique used to isolate and examine the innermost layer of the carotid artery, where endothelial cells directly contact the bloodstream. The procedure typically involves removing the vessel, opening it longitudinally, carefully separating surrounding tissue, and preserving the intimal surface for fixation, staining, or molecular analysis. In medicine and vascular research, this preparation supports en face assessment of endothelial integrity, lipid accumulation, inflammation, and atherosclerotic lesions. It enables researchers to relate structural changes in the arterial wall to vascular disease mechanisms and to evaluate treatment-related effects on carotid pathology.

This describes a partial carotid ligation surgery, which causes disturbed flow conditions and subsequent atherosclerosis development (in two weeks) with intraplaque neo-vascularization (in four weeks) in the mouse common carotid artery. We also describe a novel method of RNA isolation from the carotid intima, providing high purity endothelial RNA.

Carotid Artery and Jugular Vein Interposition: A Technique to Induce Venous Intimal Hyperplasia in Rabbit Model

This video demonstrates the interposition of the rabbit carotid artery with the jugular vein to induce venous intimal hyperplasia by subjecting the veins to arterial pressure. This model will help investigate molecular and genetic interventions to attenuate the progression of intimal hyperplasia after revascularization surgeries.

Pulse-Wave Velocity, Flow-Mediated Dilation, and Carotid Intima-Media Thickness to Assess Cardiovascular Risk in Population with Metabolic Syndrome

Endothelial/vascular aging and atherogenesis are key mechanisms that promote the development of cardiovascular diseases. The present protocol describes methods to evaluate arterial stiffness, endothelial dysfunction, and atherogenesis in patients with related risk factors, which are highly valuable in the cardiovascular research field.
