Ecm Rigidity

ECM rigidity is the mechanical stiffness of the extracellular matrix, a physical property that shapes how cells sense and respond to their tissue environment. It arises from matrix composition, collagen organization, crosslinking, and hydration, and can change during injury, fibrosis, and tumor progression. When cells attach to a rigid matrix through integrins, focal adhesions and actomyosin tension transmit mechanical signals to the nucleus, altering pathways such as YAP/TAZ and gene expression. In medicine, measuring or modulating ECM rigidity helps explain abnormal cell behavior, identify disease mechanisms, and guide drug development, biomaterial design, and regenerative therapies.
