Murine Cardiac Transplantation

Murine cardiac transplantation is a surgical model in which a donor mouse heart is implanted into a recipient to study graft function, immune responses, and transplantation biology. In the commonly used heterotopic approach, surgeons connect the donor aorta and pulmonary artery to recipient vessels, allowing the graft to be perfused while the recipient’s native heart continues circulating blood. Researchers monitor graft contractions and survival, then assess rejection, ischemia-reperfusion injury, inflammation, and immune tolerance through clinical and tissue-based analyses. This model enables controlled testing of immunosuppressive therapies and genetic or cellular interventions, providing mechanistic insight relevant to human heart transplantation and cardiovascular medicine.

The murine cervical heart transplantation model is well suited for immunological as well as ischemia reperfusion injury studies. We modified the procedure using a non-suture cuff technique and performed more than 1,000 successful transplants with this approach. Herein, we provide additional details of this technique to supplement the video.
