Transdermal Gfr Measurement

Transdermal GFR measurement is a noninvasive method for estimating glomerular filtration rate, a key indicator of kidney function, without repeated blood or urine collection. After administration of a filtration marker such as fluorescent sinistrin, a sensor placed on the skin detects changes in fluorescence as the marker is cleared from the circulation; the resulting decay curve reflects filtration kinetics. This approach can provide continuous or repeated measurements in small animals and clinical research, supporting studies of acute kidney injury, chronic kidney disease, nephrotoxic drug effects, and renal physiology. Its reduced sampling burden may improve longitudinal monitoring and experimental efficiency.

This article details the construction of a multiplexed microneedle-based sensor. The device is being developed for in situ sampling and electrochemical analysis of multiple analytes in a rapid and selective manner. We envision clinical medicine and biomedical research uses for these microneedle-based sensors.

Measurement of glomerular filtration rate (GFR) is the gold standard for kidney function assessment. Here we describe a high-throughput method which allows the determination of GFR in conscious mice by using a single bolus injection, determination of fluorescein isothiocyanate (FITC)-inulin in plasma and calculation of GFR by a two-phase exponential decay model.
