Contrast Agent Administration

Contrast agent administration is the controlled delivery of imaging agents that improve the visibility of brain structures, blood vessels, lesions, and pathological changes, making it important for neuroscience research and clinical assessment. Typically given intravenously before MRI or CT, these agents alter local magnetic relaxation or X-ray attenuation; their distribution and timing can reveal vascular perfusion, blood-brain barrier disruption, inflammation, or tumor-associated changes. Careful selection of agent, dose, imaging sequence, and monitoring supports accurate interpretation while limiting risk. The resulting contrast-enhanced images aid neuroanatomical mapping, disease characterization, treatment planning, and evaluation of therapeutic response.

Preparation and In Vitro Characterization of Dendrimer-based Contrast Agents for Magnetic Resonance Imaging

This protocol describes the preparation and characterization of a dendrimeric magnetic resonance imaging (MRI) contrast agent that carries cyclen-based macrocyclic chelates coordinating paramagnetic gadolinium ions. In a series of MRI experiments in vitro, this agent produced an amplified MRI signal when compared to the commercially available monomeric analogue.

We demonstrate how to perform macrophage MR imaging using ultrasmall superparamagnetic contrast agent (USPIO) in septic arthritis, allowing an initial and longitudinal in vivo non-invasive evaluation of macrophages infiltration and an assessment of therapy efficacy.

We demonstrate the metalation, purification, and characterization of lanthanide complexes. The complexes described here can be conjugated to macromolecules to enable tracking of these molecules using magnetic resonance imaging.
