Methylene Blue Staining

Methylene blue staining is a histological technique that uses the basic dye methylene blue to visualize cells and tissue structures under a microscope. In nervous tissue, the positively charged dye binds acidic cellular components, including nucleic acids, while its oxidation-reduction state can alter color and staining intensity. Researchers use the method to reveal neuronal cell bodies, processes, and general tissue organization, supporting studies of neuroanatomy, cellular morphology, and pathological changes. Its relatively simple workflow makes it useful for teaching microscopy and for rapid qualitative assessment of nervous system specimens.
