Microglia Signaling

Microglia signaling is the communication network through which resident immune cells of the central nervous system sense changes, coordinate responses, and influence neural function. Microglia detect damage-associated or pathogen-associated signals through cell-surface receptors, then release cytokines, chemokines, and other mediators that regulate inflammation, phagocytosis, synaptic remodeling, and interactions with neurons and glial cells. These pathways help maintain brain homeostasis and support responses to injury or infection, but persistent or dysregulated signaling can contribute to neuroinflammation and neuronal dysfunction. Studying microglia signaling in neuroscience clarifies mechanisms underlying neurodegenerative disease, brain injury, and development, while informing potential therapeutic strategies.

In this video, we describe a protocol for isolating CD11b cells, including microglia, from mouse pup brains using magnetic-activated cell sorting. The brain tissues are homogenized to obtain single-cell suspensions, which are incubated with anti-CD11b antibody-coated microbeads, followed by a magnetic column-based separation to purify the CD11b+ cell population.

We outline methods for the efficient and quick isolation/culture of viable microglia from the neonatal cerebral cortex and adult spinal cord. The dissection and plating of cortical microglia can be accomplished within 90 minutes, with the subsequent microglial harvest taking place ~ 10 days following the initial dissection.
