Ng2 Cells

NG2 cells, also called oligodendrocyte precursor cells (OPCs), are a widespread population of glial progenitors in the central nervous system that can generate myelinating oligodendrocytes and support neural circuit function. They express the NG2/CSPG4 cell-surface proteoglycan, receive synaptic input from neurons, and respond to local signals that regulate proliferation, migration, and differentiation. In neuroscience, NG2 cells provide a model for studying oligodendrocyte development, myelin formation and repair, neuron-glia communication, and responses to injury or disease. Their dynamic behavior makes them relevant to investigations of brain plasticity and potential regenerative strategies.

Here we present a novel clinical grade isolation and culture method for kidney Perivascular Stromal Cells (kPSCs) based on whole organ perfusion with digestive enzymes and NG2-cell enrichment. With this method, it is possible to acquire sufficient cell numbers for cellular therapy.

Derivation of Hematopoietic Stem Cells from Murine Embryonic Stem Cells

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

Metaphase to anaphase transition is triggered through anaphase-promoting complex (APC/C)-dependent ubiquitination and subsequent destruction of cyclin B. Here, we established a system which, following pulse-chase labeling, allows monitoring cyclin B proteolysis in entire cell populations and facilitates the detection of interference by the mitotic checkpoint.
