Oculomotor Biomarkers

Oculomotor biomarkers are measurable features of eye movements that provide noninvasive indicators of brain function and dysfunction. Eye-tracking systems quantify parameters such as saccade latency and accuracy, smooth-pursuit gain, fixation stability, and antisaccade errors as visual stimuli engage coordinated cortical, cerebellar, and brainstem circuits. In neuroscience, these measures help characterize attention, sensorimotor control, and neural connectivity, while supporting research into conditions such as Parkinson’s disease, schizophrenia, multiple sclerosis, and traumatic brain injury. Because they can be collected objectively and repeatedly, oculomotor biomarkers may improve early detection, disease monitoring, and evaluation of treatment-related changes.

Ex Vivo Culture and Imaging of Oculomotor Slices from Transgenic Mouse Embryos

This video demonstrates the ex vivo culture and imaging of oculomotor slices from transgenic mouse embryos. The embryos are isolated and embedded in agarose. Using a vibratome, slices containing the oculomotor nuclei and eyes are obtained. The slices are maintained in culture and imaged using a fluorescence microscope.

Neural control and cognitive processes can be studied through eye movements. The VisualEyes software allows an operator to program stimuli on two computer screens independently using a simple, custom scripting language. The system can stimulate tandem eye movements (saccades and smooth pursuit) or opposing eye movements (vergence) or any combination.

Biomarkers in an Animal Model for Revealing Neural, Hematologic, and Behavioral Correlates of PTSD

We describe a rat model of post traumatic stress disorder (PTSD) that reveals the persistent alterations in neuroendocrine function and the delayed long-term, exaggerated fear response, characteristic of PTSD patients. The animal model and methods described here are useful for correlating biomarkers in brain nuclei, which are mechanistic but cannot be measured in patients, with biomarkers in peripheral white blood cells, which can.

Isolation and Culture of Oculomotor, Trochlear, and Spinal Motor Neurons from a Mouse Embryo

The video demonstrates the isolation of oculomotor, trochlear, and spinal motor neurons from a transgenic mouse embryo. The embryo expresses motor neurons labeled with a green fluorescent protein (GFP) for easy visualization and identification. The motor neurons are surgically isolated under a fluorescence microscope and maintained in a culture medium.
