Oxidative Damage

Oxidative damage is the molecular injury that occurs when reactive oxygen or nitrogen species overwhelm cellular antioxidant defenses, making it an important process in neuroscience. These reactive molecules can oxidize membrane lipids, proteins, and DNA, while mitochondrial dysfunction may increase their production and intensify damage. Neurons are particularly vulnerable because of their high energy demands, abundant lipid-rich membranes, and limited regenerative capacity. Studying oxidative damage helps researchers investigate mechanisms underlying aging and neurodegenerative disorders, evaluate antioxidant and mitochondria-targeted interventions, and identify biomarkers of neuronal stress. This work also supports the development of strategies to preserve synaptic function and neuronal survival.

Assessment of Oxidative Damage in the Primary Mouse Ocular Surface Cells/Stem Cells in Response to Ultraviolet-C (UV-C) Damage

This protocol demonstrates the simultaneous detection of reactive oxygen species (ROS), live cells, and dead cells in live primary cultures from mouse ocular surface cells. 2',7'-Dichlorofluoresceindiacetate, propidium iodide, and Hoechst staining are used to assess the ROS, dead cells, and live cells, respectively, followed by imaging and analysis.

The alkaline comet assay measures DNA strand breaks in eukaryotic cells. By adding an Endonuclease III or human 8-oxoguanine-DNA-N-glycosylase digestion step, the assay can efficiently detect oxidative DNA damage. We describe methods for using these assays to detect DNA damage in rat liver.
