Sclera Choroid Rpe Dissection

Sclera, choroid, and RPE dissection is a microsurgical technique that separates three anatomically distinct layers of the eye for structural and molecular analysis in neuroscience. Under magnification, the tissue is carefully opened and peeled apart along natural interfaces, allowing the fibrous sclera, vascular choroid, and pigmented retinal pigment epithelium to be isolated while minimizing damage and cross-contamination. The resulting layer-specific samples support histological examination, gene and protein analysis, and investigation of retinal and choroidal disorders. This approach helps researchers relate cellular changes in the ocular tissues to visual processing, neurodegeneration, inflammation, and disease progression.

The human retina is composed of functionally and molecularly distinct regions, including the fovea, macula, and peripheral retina. Here, we describe a method using punch biopsies and manual removal of tissue layers from a human eye to dissect and collect these distinct retinal regions for downstream proteomic analysis.

Choroid Sprouting Assay to Study Ocular Microvascular Proliferation Ex Vivo

2025

This video demonstrates the development of an ex vivo model of choroidal sprouting to study ocular microvascular angiogenesis. The assay involves culturing mouse eye explants containing the choroid tissue attached to the sclera and retinal pigment epithelium under specific conditions that promote choroidal microvascular sprouting.

This video presentation shows a method of harvesting the two most important highly vascular structures that support forebrain function. They are the cerebral surface (superficial) vasculature along with associated meninges (MAV) and the choroid plexus which are necessary for cerebral blood flow and cerebrospinal fluid (CSF) homeostasis.
