Single Fiber Emg

Single-fiber electromyography (SFEMG) is a specialized neurophysiological technique that records the electrical activity of individual muscle fibers, providing highly sensitive information about neuromuscular function. A fine needle electrode detects action potentials from nearby fibers during voluntary contraction or stimulation, allowing measurement of neuromuscular jitter, the variation in time between fiber discharges, and fiber density. In neuroscience and clinical neurophysiology, these measurements help evaluate neuromuscular transmission and identify abnormalities associated with disorders such as myasthenia gravis and other motor-unit diseases. SFEMG complements nerve conduction studies and conventional EMG by revealing subtle dysfunction that broader recordings may miss.

In Vivo Single-Fiber Recording of Sciatic Nerve C-Fibers in Rats

This video demonstrates a single-fiber recording of C-fiber nerve conduction in rats. An anesthetized rat is incised to expose the sciatic nerve, which provides sensory and motor supply to the lower limbs. The individual C-fibers are then exposed. Electrical stimulation is provided using stimulus electrodes implanted in the foot, and a response is recorded from a single nerve fiber.

Extraction of the EPP Component from the Surface EMG

The endplate potential (EPP) component can be extracted from the surface EMG using a digital filter. The extracted EPP shows oscillation with a frequency of about 30 Hz.

Here we present a novel experimental protocol and data collection technique for simultaneous non-invasive acquisition of neural activity, muscle activity, and whole-body kinematics during various locomotion tasks and conditions.
