Tumor Morphology

Tumor morphology is the study of a tumor’s size, shape, cellular organization, and tissue architecture, providing clues about its identity and biological behavior. In neuroscience, researchers assess morphology through neurological imaging and microscopic examination of stained tissue, evaluating features such as cellular arrangement, boundaries, vascular patterns, and invasion into surrounding brain regions. These structural characteristics help distinguish tumor types, support diagnosis, and indicate how a lesion may affect neural circuits and brain function. Morphological analysis also guides tumor classification, treatment planning, and research into how abnormal growth, tissue infiltration, and interactions with the nervous system influence disease progression.

Our protocol describes how to dissect the rat abdominal mammary gland and how to prepare mammary gland whole mounts. It also describes how to analyze mammary gland morphology using three end-points (number of terminal end buds, epithelial elongation and differentiation) and to use these results to predict mammary cancer risk in rats which were exposed to dietary modifications in utero or during prepuberty.

Optical coherence tomography (OCT), a three-dimensional imaging technology, was used to monitor and characterize the growth kinetics of multicellular tumor spheroids. Precise volumetric quantification of tumor spheroids using a voxel counting approach, and label-free dead tissue detection in the spheroids based on intrinsic optical attenuation contrast, were demonstrated.
