Mood Fluctuations

Mood fluctuations are changes in a person’s emotional state over time, ranging from brief shifts in feelings and energy to more persistent variations that may affect daily functioning. They occur as cognitive appraisal, physiological arousal, and responses to internal or environmental cues interact, with factors such as stress, sleep, and social experiences influencing the intensity and duration of mood changes. In psychology, examining these patterns helps distinguish typical emotional variation from clinically significant symptoms, supports assessment of mood disorders, and informs interventions that target coping, emotional regulation, and well-being.

This video demonstrates the technique of fluorescence fluctuation spectroscopy (FFS) to study protein homo-oligomerization. Fluorescently-labeled proteins in a sample are dimerized using an agent when studying protein oligomerization using FFS. Using a confocal microscope, as the proteins move in and out of the small observation volume, the analysis of the fluctuations in the brightness of the fluorescent molecules is performed to determine the oligomeric state of the proteins.

Here, a protocol is presented to perform a fluctuation assay and estimate microbial mutation rate using phenotypic markers. This protocol will enable researchers to assay mutations in diverse microbes and environments, determining how genotype and ecological context affect spontaneous mutation rates.
