Transgenic Plants

Le piante transgeniche sono prodotte introducendo un gene estraneo nel genoma di una pianta. Queste piante possono essere sviluppate per una serie di motivi, tra cui la resistenza a virus o parassiti, una resa più elevata o la produzione di proteine terapeutiche.
Per produrre una pianta transgenica, un gene di interesse e il suo promotore vengono introdotti nel protoplasto vegetale, una cellula vegetale priva di parete cellulare, incubando entrambi in un terreno contenente polietilenglicole.
Il protoplasto ricombinante viene coltivato su un terreno sterile in presenza di regolatori di crescita e sostanze nutritive.
Le cellule del protoplasto crescono indefinitamente, producendo una massa di cellule indifferenziate chiamata callo. Alcune cellule del callo, chiamate cellule totipotenti, danno origine a nuovi germogli e radici, che possono svilupparsi in una pianta completamente nuova.
Le piante transgeniche possono anche essere create utilizzando il batterio Agrobacterium tumefaciens, un batterio gram-negativo che si trova nel suolo. Questo batterio è un agente patogeno che normalmente causa malattie nelle piante e contiene un plasmide Ti che può integrarsi nel genoma di una cellula vegetale.
Il plasmide Ti ha due regioni di interesse: il T-DNA o la regione del DNA di trasferimento che può essere inserita nel genoma della cellula ospite e la regione di virulenza per favorire il trasferimento del T-DNA nella cellula ospite.
Questo plasmide viene modificato inserendo un gene desiderato nella regione del T-DNA e un gene per la resistenza agli antibiotici per consentire la crescita selettiva di organismi contenenti plasmidi. Il plasmide ricombinante viene quindi inserito nei batteri applicando un impulso elettrico per aumentare la permeabilità cellulare e consentire l'assorbimento del plasmide nei batteri.
L'Agrobacterium tumefaciens che trasporta il plasmide ricombinante viene coltivato in un terreno di coltura contenente antibiotici.
Le parti della pianta, come i nodi cotiledonari di una pianta di pomodoro, vengono tagliate e incubate nel terreno contenente i batteri.
Le cellule vegetali ferite sul bordo del nodo staminale rilasciano composti che attirano i batteri. Il batterio infetta la pianta e rilascia il plasmide ricombinante, che poi trasferisce il gene estraneo nel genoma della cellula vegetale.
Solo le cellule trasformate con DNA ricombinante e geni resistenti agli antibiotici sopravvivono nel terreno contenente antibiotici.
Le cellule trasformate ora formano un callo e si sviluppano in una nuova pianta transgenica che porta un transgene.

La tecnologia del DNA ricombinante chiamata transgenesi viene spesso utilizzata per aggiungere un gene estraneo o rimuovere un gene dannoso da un organismo. Tali organismi geneticamente modificati sono chiamati organismi transgenici.

La prima pianta transgenica è stata una pianta di tabacco sviluppata nel 1983 che ha mostrato resistenza al virus del mosaico del tabacco. Da allora, molte piante transgeniche sono state sviluppate e commercializzate per migliorare il valore agricolo, ornamentale e orticolo di una pianta coltivata. Le piante transgeniche possono anche essere utilizzate come bioreattore per la produzione di metaboliti o proteine economicamente importanti.

Metodi fisici per trasferire i geni alle piante

Molti metodi, come l'elettroporazione e il bombardamento di microproiettili, possono essere utilizzati per trasferire un gene estraneo in un protoplasto vegetale. Durante l'elettroporazione, un dispositivo fornisce impulsi ad alta tensione alla sospensione del protoplasto. Ciò aumenta la permeabilità del protoplasto consentendo al DNA estraneo di entrare nella cellula e integrarsi nel genoma della pianta.

Al contrario, durante il bombardamento di microproiettili o il metodo biolistico, le particelle sferiche di tungsteno o oro (microproiettili) vengono rivestite con il gene che trasporta il plasmide di interesse. Le particelle rivestite vengono quindi accelerate a una velocità compresa tra 300 e 600 m/s con uno strumento chiamato pistola a particelle. Lo strumento utilizza gas elio ad alta pressione per fornire forza propulsiva che aiuta le particelle rivestite di plasmide a penetrare nella parete cellulare della pianta. Una volta all'interno della cellula, il gene di interesse si integra nel genoma della pianta. Questo metodo può essere utilizzato per introdurre DNA in colture di calli, tessuti meristematici, embrioni immaturi, sospensioni di cellule vegetali, ecc.

Applicazioni delle piante transgeniche

La tecnologia del DNA ricombinante è ampiamente utilizzata nello sviluppo di piante resistenti ai parassiti e ai virus. La tecnica è utile anche per modificare il contenuto di lipidi, amido e proteine dei semi.

Ad esempio, il riso dorato è una pianta di riso geneticamente modificata con un valore nutritivo migliorato. Questa varietà transgenica di riso produce β-carotene, un precursore della vitamina A, che gli conferisce un colore dorato. È prodotto dalla trasformazione del DNA mediata da Agrobacterium, in cui il DNA ricombinante che trasporta tre geni che codificano gli enzimi fitoene sintasi, fitoene desaturasi e licopene β-ciclasi viene inserito nel genoma degli embrioni di riso bersaglio. Gli embrioni si sviluppano poi in piante che producono β-carotene. Il riso dorato è stato sviluppato per contrastare la carenza di vitamina A nei paesi in via di sviluppo.

Alcune altre piante transgeniche, come il cotone Bt, sono state modificate inserendo un gene isolato dal batterio Bacillus thuringiensis, un batterio gram-positivo presente nel suolo. Tali piante transgeniche producono proteine cristalline o “Cry” che uccidono gli insetti dannosi, rendendo la pianta resistente agli insetti.

Transgenic plants are crops that have been changed with recombinant DNA technology, also called transgenesis. This method adds a foreign gene or removes a harmful one from an organism. The result is a genetically modified organism. The first transgenic plant was a tobacco plant made in 1983, and it was resistant to tobacco mosaic virus.

Since then, transgenic plants have been developed for many uses in agriculture, horticulture, and ornamental plants. Some are designed to resist pests or viruses. Others are used to improve the lipid, starch, or protein content of seeds. In some cases, transgenic plants can also act as bioreactors and make useful metabolites or proteins.

Foreign genes can be transferred into plant protoplasts by physical methods such as electroporation and microprojectile bombardment. In electroporation, high-voltage pulses make the protoplast membrane more permeable. This allows the foreign DNA to enter the cell and integrate into the plant genome. In microprojectile bombardment, also called the biolistic method, tungsten or gold particles are coated with plasmid DNA and shot into the cells with a particle gun.

The particle gun uses high-pressure helium gas to accelerate the coated microprojectiles to about 300 to 600 m/s. The particles can penetrate the plant cell wall and deliver the gene of interest. This method can be used with callus cultures, meristematic tissues, immature embryos, and plant cell suspensions. Once inside the cell, the introduced DNA can integrate into the plant genome.

A well-known example is golden rice, which has enhanced nutritional value. It produces β-carotene, a precursor of vitamin A, and this gives the rice its golden color. Golden rice was made by Agrobacterium-mediated DNA transformation using recombinant DNA with genes for phytoene synthase, phytoene desaturase, and lycopene β-cyclase. It was developed to help address vitamin A deficiency in developing nations.

Another example is Bt cotton. It contains a gene from Bacillus thuringiensis, a gram-positive soil bacterium. The plant makes Cry protein, which kills harmful insects and makes the crop insect resistant. These examples show how transgenic plants can improve crop value and help solve real agricultural problems.
