La terapia genica &#xE8; una tecnica in cui un gene viene inserito nelle cellule di una persona per prevenire o trattare una malattia grave. Il gene aggiunto pu&#xF2; essere una versione sana del gene che &#xE8; mutato nel paziente, o potrebbe essere un gene diverso che inattiva o compensa il gene che causa la malattia del paziente. Ad esempio, nei pazienti con immunodeficienza combinata grave (SCID) a causa di una mutazione nel gene per la deaminasi enzimatica adenosina, pu&#xF2; essere inserita una versione funzionante del gene. Le cellule del paziente possono quindi rendere l&#x27;enzima, curando questa malattia potenzialmente mortale in alcuni casi.

<h4>Inserimento genico</h4>

I geni possono essere introdotti nelle cellule di un paziente in due modi principali: in vivo, direttamente in una persona attraverso l&#x27;iniezione in tessuti specifici o nel flusso sanguigno; ed ex vivo, nelle cellule che sono state rimosse dal paziente, che vengono trapiantate dopo l&#x27;inserimento del gene.

Il gene viene solitamente inserito in un vettore, spesso un virus che &#xE8; stato modificato per non causare la malattia, per ottenere il gene nelle cellule del paziente e consegnato al nucleo. In alcuni casi, ad esempio quando si utilizzano vettori retrovirali, il gene viene inserito casualmente nel genoma della persona, portando all&#x27;espressione stabile del gene inserito. In altri, come quando si utilizzano vettori adenovirali, il gene non si integra nel genoma dell&#x27;ospite ed &#xE8; espresso solo in modo transitorio.
