Allosteric Proteins-ATCase

Le proteine che contengono siti di legame collegati sono chiamate proteine allosteriche e possono essere di vario tipo, come enzimi, recettori, proteine strutturali e proteine motorie. 
Ad esempio, l'aspartato transcarbamilasi, nota anche come ATCasi, è un grande enzima contenente sei subunità catalitiche, ciascuna con un sito di legame del substrato. Ha anche sei subunità regolatorie aggiuntive, ciascuna con un sito in grado di legare sia le pirimidine che le purine, i mattoni del DNA e dell'RNA.
L'enzima catalizza un passaggio essenziale nella sintesi della pirimidina, la condensazione dell'aspartato e del carbamil fosfato per produrre carbamil aspartato. La velocità di questa reazione aumenta in modo dipendente dalla concentrazione.
Il legame dell'ATP, una purina, all'enzima attiva l'enzima. Mentre, il legame simultaneo di UTP e CTP, le pirimidine, si traduce in un'inibizione del 95% dell'attività enzimatica.

I collegamenti ai siti di legame possono regolare la funzione di una proteina. Ad esempio, l’attività enzimatica è spesso regolata attraverso un meccanismo di feedback in cui il prodotto finale del processo biochimico funge da inibitore.

L'aspartato transcarbamilasi(ATCase) è un enzima citosolico che catalizza la condensazione di L-aspartato e carbamoil fosfato in N-carbamoil-L-aspartato. Questa reazione è il primo passo nella biosintesi della pirimidina. UTP e CTP, i prodotti finali della via di sintesi della pirimidina, inibiscono l'attività di ATCase, l'enzima che catalizza il primo passaggio essenziale di questa via. Il legame di UTP e CTP all'enzima regola negativamente il sito catalitico collegato quando la concentrazione di pirimidine è elevata, rispetto alla concentrazione di purine nella cellula. ATCase fa parte del complesso multienzimatico CAD, parte della via di biosintesi della pirimidina, insieme alla carbamoil fosfato sintetasi II e alla diidroorotasi. Le pirimidine sono essenziali per la sintesi del DNA durante la divisione cellulare, pertanto l'inibizione dell'attività della ATCase rallenta la crescita del tumore nel cancro.

ATCase is a key enzyme in pyrimidine synthesis. It is a cytosolic enzyme that catalyzes the condensation of L-aspartate and carbamoyl phosphate to form N-carbamoyl-L-aspartate. This reaction starts the pathway that makes pyrimidines.

ATCase is regulated by feedback inhibition. The end products UTP and CTP bind to the enzyme and reduce its activity when pyrimidine levels are high compared with purines in the cell. This binding at one site affects the linked catalytic site, which helps keep metabolite levels balanced.

ATCase also works as part of the CAD multi-enzyme complex. CAD includes carbamoyl phosphate synthetase II and dihydroorotase in the pyrimidine biosynthesis pathway. Because pyrimidines are needed for DNA synthesis during cell division, blocking ATCase activity can slow tumor growth in cancer.
