Fluorescence Activation

Fluorescence activation is the conversion of a nonfluorescent or weakly fluorescent label into a detectable fluorescent state, enabling researchers to track biochemical molecules and events with high spatial and temporal resolution. Activation can occur when light or a chemical reaction alters the fluorophore’s structure, removes a fluorescence-quenching group, or exposes a previously hidden fluorescent component. In biochemistry, this principle supports signal amplification, molecular localization, reaction monitoring, and activity-based assays. By restricting fluorescence to selected molecules or time points, fluorescence activation can reduce background signals and help clarify protein interactions, enzyme function, transport, and dynamic cellular processes.

Detection of Inflammasome Activation via Fluorescence Microscopy

This video demonstrates a method to detect NLRP3 inflammasome activation in macrophages using fluorescent reporter probes. The cells are treated with potassium ionophores and glycine to induce inflammasome assembly and caspase-1 activation. Fluorescent reporter probes visualize activated caspase-1, and cells are stained with a fluorescent dye for nuclei visualization.

A method for isolating specific cell types from plant material is demonstrated. This technique employs transgenic marker lines expressing fluorescent proteins in particular cell types, cellular dissociation and Fluorescence Activated Cell Sorting. Additionally, a growth setup is established here that facilitates treatment of Arabidopsis thaliana seedlings prior to cell sorting.

The Protease Fluorescent Detection Kit is designed for the measurement of protease activity using fluorometry. It is also suitable for detection of trace amounts of protease contamination. The method is based on the proteolytic hydroysis of a proprietary formulation of a FITC-labeled casein substrate.
