Nbd-c2 Lact

NBD-C2 Lact is a fluorescent lactosylceramide analog used to study sphingolipid behavior in biochemical and cellular systems. Its short C2 acyl chain improves experimental handling, while the attached nitrobenzoxadiazole (NBD) fluorophore produces a detectable signal when illuminated at appropriate wavelengths; changes in fluorescence can indicate the probe’s membrane environment, localization, or transfer. Researchers use NBD-C2 Lact to examine lipid trafficking, membrane organization, and glycosphingolipid metabolism, including movement through secretory and endocytic pathways. As a labeled lipid substrate, it supports visualization and quantitative analysis of processes that are difficult to measure using unlabeled lactosylceramide.

Here, we present a protocol to detect single, SNARE-mediated fusion events between liposomes and supported bilayers in microfluidic channels using polarized TIRFM, with single molecule sensitivity and ~15 msec time resolution. Lipid and soluble cargo release can be detected simultaneously. Liposome size, lipid diffusivity, and fusion pore properties are measured.
