Single Molecule Studies

Single molecule studies are experimental approaches that examine the behavior of individual molecules, revealing molecular heterogeneity and transient events that ensemble measurements can obscure. In biochemistry, researchers track or manipulate single proteins, nucleic acids, or molecular complexes using methods such as fluorescence microscopy, optical tweezers, force spectroscopy, and nanopore sensing; these techniques detect signals from individual molecules as they bind, move, fold, or undergo conformational changes. The resulting measurements connect molecular structure with kinetics and mechanical properties, clarify how biomolecular machines function, and improve understanding of processes such as enzyme catalysis, protein folding, and nucleic acid dynamics.

Surface Passivation for Single-molecule Protein Studies

We describe a method for passivating a glass surface using polyethylene glycol (PEG). This protocol covers surface cleaning, surface functionalization, and PEG coating. We introduce a new strategy for treating the surface with PEG molecules over two rounds, which yields superior quality of passivation compared to existing methods.
