Tmt Labeling

TMT labeling is a chemical method for comparing protein abundance across biological samples using tandem mass spectrometry. Amine-reactive tandem mass tags attach to peptide N termini and lysine side chains, creating isobaric derivatives that co-elute and appear as a single precursor during the initial mass-spectrometry scan; fragmentation releases sample-specific reporter ions whose intensities reflect relative peptide abundance. By labeling multiple samples in one experiment, researchers can perform multiplexed quantitative proteomics while reducing instrument time and technical variation. The approach supports studies of cellular pathways, disease-associated protein changes, post-translational modifications, and responses to experimental treatments.
