Automated Sample Centering

Automated sample centering is a positioning technique that locates and aligns a specimen within an imaging or measurement system, improving consistency and reducing manual adjustment. It typically combines optical or image-based detection with software-controlled motorized stages, which identify the sample’s geometry or signal, calculate its center, and move the specimen into a defined measurement position. In bioengineering, this process supports microscopy, microfluidics, biomaterials analysis, and automated experimental workflows by increasing measurement repeatability, reducing operator bias, and accelerating sample throughput. Reliable centering also helps standardize data collection across specimens, enabling more efficient characterization of cells, tissues, and engineered biological systems.

Here, we present an automated plasma protein digestion method for mass spectrometry-based quantitative proteomic analysis. In this protocol, the liquid transferring and incubation steps for protein denaturation, reduction, alkylation, and trypsin digestion reactions are streamlined and automated.
