Cell Membrane Potential

Cell membrane potential is the voltage difference across a cell membrane created by unequal distributions of ions and selective membrane permeability; it is central to how cells sense and transmit signals. In living cells, ion pumps such as the sodium-potassium pump establish concentration gradients, while ion channels open or close to regulate ionic flow and rapidly change the membrane voltage. In bioengineering, measuring and controlling membrane potential supports electrophysiology, neural interfaces, biosensors, and engineered tissues. These approaches help characterize excitable cells, evaluate the effects of drugs or stimulation, and guide the design of devices that communicate with biological systems.

Mitochondria can utilize the electrochemical potential across their inner membrane (ΔΨm) to sequester calcium (Ca2+), allowing them to shape cytosolic Ca2+ signaling within the cell. We describe a method for simultaneously measuring mitochondria Ca2+ uptake and ΔΨm in live cells using fluorescent dyes and confocal microscopy.
