Hollow Fiber Bioreactor

A hollow fiber bioreactor is a three-dimensional cell-culture system that uses bundles of semipermeable, tube-shaped fibers to support high-density cell growth under controlled conditions. Culture medium flows through the fiber lumens or around the fiber surfaces, allowing nutrients and oxygen to diffuse across the membranes while metabolic wastes move away from the cell compartment. This perfusion-based design provides a large surface area for mass transfer and can maintain cells in environments that more closely resemble tissue than conventional two-dimensional culture. In bioengineering, hollow fiber bioreactors support production of biologics, tissue-engineering research, and studies requiring prolonged, scalable cell cultivation.

This protocol describes a method for producing extracellular vesicles at high concentrations on a continuous basis while controlling cell density in hollow fiber bioreactors.

Production of Extracellular Matrix Fibers via Sacrificial Hollow Fiber Membrane Cell Culture

The goal of this protocol is production of whole extracellular matrix fibers targeted for wound repair which are suitable for preclinical evaluation as part of a regenerative scaffold implant. These fibers are produced by the culture of fibroblasts on hollow fiber membranes and extracted by dissolution of the membranes.
