Silicone Mold Casting

Silicone mold casting is a fabrication method in which a liquid or uncured material is shaped inside a flexible silicone mold, allowing accurate replication of a master geometry. Typically, silicone elastomer is mixed with a curing agent, poured around a patterned master, degassed to reduce trapped bubbles, and cured before the mold is removed and filled with another material such as a hydrogel, polymer, or resin. In bioengineering, this approach supports rapid prototyping of microfluidic devices, tissue-engineering scaffolds, anatomical models, and laboratory tools with complex or delicate features. Reusable molds and low-temperature processing can also help preserve sensitive biomaterials.

2025

This protocol presents an improved demolding procedure for the Polydimethylsiloxane (PDMS) double casting technique by introducing silicone oil as a non-adhesive barrier between PDMS layers. Unlike plasma or chemical surface modification methods, this approach requires no specialized or costly equipment, making it a more accessible and cost-effective alternative.

This article introduces a unique method for fabricating master molds with an air-vent hole to prevent air entrapment during viscous polymer casting, enabling the fabrication of gelatin microneedles with sharp tips. By reducing the casting volume of high-viscosity gelatin, thin-walled hollow pyramidal microneedles could also be fabricated.
