Smooth Muscle Contractility

Smooth muscle contractility is the ability of involuntary muscle cells to generate and regulate force, a property that controls the diameter and movement of structures such as blood vessels, airways, and the gastrointestinal tract. Contractile stimuli raise intracellular calcium, which binds calmodulin and activates myosin light-chain kinase; phosphorylation of myosin enables actin-myosin cross-bridge cycling, while calcium removal promotes relaxation. In bioengineering, measuring smooth muscle contractility helps evaluate engineered tissues, model vascular and airway function, and assess how cells respond to mechanical or biochemical cues. These studies support the design of physiological tissue models and strategies for treating contractile dysfunction.

Ex Vivo Assessment of Contractility, Fatigability and Alternans in Isolated Skeletal Muscles

We describe a method to directly measure muscle force, muscle power, contractile kinetics and fatigability of isolated skeletal muscles in an in vitro system using field stimulation. Valuable information on Ca2+ handling properties and contractile machinery of the muscle can be obtained using different stimulating protocols.
