Methylcellulose Overlay

Methylcellulose overlay is a biological technique that uses a viscous, semisolid layer to control the movement of cells, particles, or soluble factors across a culture surface. In virological plaque assays, the methylcellulose matrix limits diffusion of newly produced virions, so infection remains localized and expands through neighboring cells rather than spreading freely through the medium. This spatial restriction produces distinct zones of cell damage, or plaques, that researchers can count to estimate infectious virus concentration. Methylcellulose overlays are therefore valuable for measuring viral infectivity, comparing replication phenotypes, and studying cell-to-cell transmission under controlled culture conditions.

Plaque assays are the gold standard for viral quantification, utilizing entrapping overlays on host cellular monolayers to determine viral titers. While various semisolid overlays have traditionally been used, here we demonstrate plaque techniques comparing semisolid overlays to a novel liquid microcrystalline cellulose among several families of viruses.

Testing protein-protein interaction is indispensable for dissection of protein functionality. Here, we introduce an in vitro protein-protein binding assay to probe a membrane-immobilized protein with a soluble protein. This assay provides a reliable method to test interaction between an insoluble protein and a protein in solution.
