Ros Quantification

ROS quantification is the measurement of reactive oxygen species in biological samples, providing an important way to assess cellular redox balance and oxidative stress. In common assays, chemical or genetically encoded probes react with specific ROS or undergo oxidation, producing a fluorescence, absorbance, or luminescence signal that is compared with controls and calibrated measurements. This technique supports studies of mitochondrial function, inflammation, toxicology, aging, and disease mechanisms by revealing changes in ROS production and antioxidant responses. Careful probe selection, sampling conditions, and normalization are essential because ROS are short-lived and highly reactive.

2023

This video describes the protocol for detecting total cellular reactive oxygen species (ROS) using 2',7'-dichlorodihydrofluorescein diacetate (DCFH-DA). This method helps visualize ROS localization in the adherent cells using a fluorescence microscope and further quantify ROS intensity with a fluorescence plate reader.

This protocol describes the use of a genetically encoded hydrogen peroxide (H2O2)-biosensor in cultured zebrafish neurons and larvae for assessing the physiological signaling roles of H2O2 during nervous system development. It can be applied to different cell types and modified with experimental treatments to study reactive oxygen species (ROS) in general development.
