Organotypic Brain Slices

Organotypic brain slices are thin sections of brain tissue maintained in vitro while preserving much of the native cellular organization, synaptic connectivity, and local microenvironment. Typically prepared from viable tissue and cultured at an air–liquid interface or submerged in nutrient medium, these slices support neuronal and glial survival through controlled temperature, nutrients, and gas exchange while allowing experimental access to defined brain regions. In biological research, organotypic brain slices provide a bridge between cell culture and animal studies for investigating neural circuits, synaptic plasticity, neurodevelopment, injury, disease mechanisms, and responses to pharmacological treatments. Their accessible architecture also supports imaging, electrophysiology, and targeted molecular manipulation.

Coculture System with an Organotypic Brain Slice and 3D Spheroid of Carcinoma Cells

The organotypic brain slice coculture with carcinoma cells enables visualizing morphological changes by fluorescence as well as bright field (video) microscopy during the process of carcinoma cell invasion of brain tissue. This model system also allows for cell exchange and replenishment approaches and offers a wide variety of manipulations and analyses.

We describe a method to prepare organotypic hippocampal slices that can be easily adapted to other brain regions. Brain slices are laid on porous membranes and culture media is allowed to form an interface. This method preserves the gross architecture of the hippocampus for up to 2 weeks in culture.
