Viable Tissue Differentiation

Viable tissue differentiation is the process of guiding living, viable cells within a tissue or culture system toward specialized cell types while preserving their structure and function. It occurs when biochemical signals, growth factors, extracellular matrix cues, cell-cell interactions, and culture conditions regulate gene expression and cellular maturation. In biological techniques, researchers assess differentiation through changes in morphology, phenotype, viability, and tissue-specific function. This approach supports tissue engineering, regenerative medicine, disease modeling, and drug testing by producing physiologically relevant cell populations and tissue constructs for studying development, injury, and therapeutic responses.

Pancreatic metaplastic cells are precursors of malignant cells that give rise to pancreatic tumors. However, isolating intact viable pancreatic cells is challenging. Here, we present an efficient method for pancreatic tissue dissociation. The cells can then be used for single-cell RNA sequencing (scRNA-seq) or for two- or three-dimensional co-culturing.

A method for rapid isolation of mitochondria from mammalian tissue biopsies is described. Rat liver or skeletal muscle preparations were homogenized with a commercial tissue dissociator and mitochondria were isolated by differential filtration through nylon mesh filters. Mitochondrial isolation time is <30 min compared to 60 - 100 min using alternative methods.
