C. Elegans Lipid Metabolism

C. elegans lipid metabolism is the network of processes that acquire, synthesize, store, transport, and break down fats in the nematode, making it a useful model for studying energy balance and metabolic health. Dietary lipids are processed primarily in the intestine, converted into neutral fats, and stored in lipid droplets; when energy demands change, metabolic enzymes mobilize these reserves under regulation from nutrient-sensing pathways such as insulin-like signaling. Researchers use C. elegans to investigate lipid storage, obesity-related mechanisms, aging, reproduction, and metabolic disease, often by combining genetic manipulation with fluorescence-based measurements of fat distribution and abundance.

Overview of Lipid Metabolism

2024

Lipid metabolism is a crucial process in the human body that involves the synthesis and degradation of lipids. This process is essential for energy production, cell membrane formation, and hormone production, among other functions. Lipolysis: The Breakdown of Lipids: Lipolysis is the process of breaking down lipids, particularly triglycerides, into glycerol and fatty acids. This process typically occurs in the adipose tissue and is triggered by various hormones, including glucagon and...

The easiness of maintaining and propagating the nematode C. elegans make it a nice model organism to work with. The possibility of synchronizing worms allows the work with a significant amount of subjects at the same developmental stage, what facilitates the study of one particular process in many animals.

Zebrafish represent a powerful vertebrate model that has been under-utilised for metabolic studies. Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.

Nile Red Staining of C. elegans: A Method to Visualize Lipid Droplets in Fixed Animals

The food source for Caenorhabditis elegans in the lab is live Escherichia coli. Since bacteria are metabolically active, they present a confounding variable in metabolic and drug studies in C. elegans. A detailed protocol to metabolically inactivate bacteria using paraformaldehyde is described here.
