Tubulin Expression

Tubulin expression is the cellular production and regulation of tubulin proteins, the building blocks of microtubules that organize cell structure and movement. It begins with transcription of tubulin genes and translation of their messenger RNAs, followed by formation of α/β-tubulin heterodimers that assemble into dynamic microtubule polymers; cells adjust expression and assembly in response to developmental and physiological demands. Measuring tubulin expression with methods such as quantitative PCR, immunoblotting, or immunostaining helps researchers assess cytoskeletal organization, cell division, neuronal development, and responses to microtubule-targeting drugs. These analyses support studies of basic cell biology and diseases involving altered cytoskeletal regulation.

This protocol describes a technique for the high-yield isolation of tubulin from the porcine brain optimized for small-scale instrumentation. The isolation procedures are complemented by procedures for determining tubulin polymerization activity in vitro using co-sedimentation assays and transmission electron microscopy.

This procedure describes a quick and easy workflow to introduce siRNA into difficult to transfect cell lines and follow gene expression by real-time PCR. Use of an automated cell counter, multi-well electroporation plate, and automated electrophoresis station provide quick and reliable results without the need for expensive robotic handling.
