Cell Enrichment

Cell enrichment is the process of increasing the proportion of a desired cell type within a mixed biological sample, making downstream analysis or experimentation more focused and reliable. It works by exploiting differences among cells, such as surface markers, size, density, adhesion, or physical properties, using approaches that may include antibody-based magnetic separation, flow sorting, density-gradient centrifugation, or selective culture conditions. In biology, enriched populations support cell characterization, molecular profiling, disease research, stem cell studies, and therapeutic development. The quality of enrichment depends on recovery, purity, cell viability, and the degree to which the procedure preserves native cell behavior.

The goal of this protocol is to manufacture pathogen-specific clinical-grade T cells using a bench-top, automated, second generation cell enrichment device that incorporates a closed cytokine capture system and does not require dedicated staff or use of a GMP facility. The cytomegalovirus pp65-specific-T cells generated can be directly administered to patients.
