Cryovial Recovery Method

Cryovial Recovery Method is a laboratory procedure used to revive biological samples preserved at cryogenic temperatures, restoring cells or microorganisms to viable, growing cultures for biological research. Recovery typically involves rapidly thawing a sealed cryovial in a controlled warm environment, transferring its contents into suitable growth medium, and providing conditions that support cell survival while reducing exposure to concentrated cryoprotectant. Careful handling during thawing, dilution, centrifugation when appropriate, and initial incubation helps limit osmotic or thermal stress and contamination. The method enables researchers to preserve valuable strains, establish consistent starting cultures, and resume experiments involving cell biology, microbiology, biobanking, and downstream analyses.

This protocol describes a procedure to freeze muscle tissues by plunging them directly into liquid nitrogen. This protocol also highlights a new cryovial that can avoid the "blanket effect" of nitrogen gas when liquid nitrogen contacts the tissue surface of a specimen.

Preparation of Brain Sections Using a NMDG-Based Protective Recovery Method

2025

This video describes a protocol to obtain brain slices using an N-Methyl-D-glucamine (NMDG)-based protective recovery method. A dissected brain is incubated in an aCSF with NMDG and HEPES to minimize neuronal cell death and damage during the sectioning. The section is allowed to recover in the aCSF to restore the cellular...

(2017)This video demonstrates the recovery of infectious Zika virus using electroporation of in vitro transcribed RNA. The process shows how synthetic RNA initiates infection and leads to viral replication in cultured cells.
