Culture Activation

Culture activation is the process of stimulating a biological culture so its cells or microorganisms resume metabolic activity, growth, or a defined functional response under controlled laboratory conditions. Activation typically involves providing suitable nutrients, growth factors, environmental conditions, or experimental stimuli that engage cellular signaling, gene expression, and proliferation, depending on the culture system. In biology, this step helps establish healthy cell cultures, initiate microbial growth, and prepare samples for downstream experiments. Consistent activation improves culture performance and reproducibility, supporting research in cell biology, microbiology, disease modeling, biomanufacturing, and the development of culture-based assays.

2011

A method for developing cell culture substrates with the ability to change topography during culture is described. The method makes use of smart materials known as shape memory polymers that have the ability to memorize a permanent shape. This concept is adaptable to a wide range of materials and applications.

Recording Electrophysiological Activity of the Neuronal Network in a Neuron-Astrocyte Co-culture

2025

This video demonstrates establishing a neuron-astrocyte co-culture for recording neuronal network activity. Upon establishing the co-culture of motor neurons and astrocytes on a multi-electrode array (MEA) plate, the synchronous rhythmic electrical activity of the neuronal network was recorded extracellularly using the MEA.

Culturing of Activated T Cells from Human Peripheral Blood Mononuclear Cells

In this video, we describe a protocol for human primary T cell activation. The incubation of naïve T cells with magnetic beads coated with anti-CD3 and anti-CD28 antibodies, followed by interleukin-2 or interleukin-15 treatment, leads to the activation of naïve T cells and their differentiation and proliferation into effector or memory T cells.

Neuron-Macrophage Co-cultures to Activate Macrophages Secreting Molecular Factors with Neurite Outgrowth Activity

The current protocol presents experimental procedures to stimulate cultured macrophages to be endowed with capacity to release molecular factors that promote neurite outgrowth. Treatment of cAMP to the neuron-macrophage co-cultures induces the macrophages to produce conditioned medium that possesses strong neurite outgrowth activity.

This protocol details an investigation of the early interactions between virally infected nasal epithelial cells and innate cell activation. Individual subsets of immune cells can be distinguished based on their activation in response to viral infections. They can then be further investigated to determine their effects on early antiviral responses.
