Epithelium Isolation

Epithelium isolation is a laboratory method for separating epithelial tissue or epithelial cells from surrounding tissues so they can be examined, cultured, or analyzed independently. The process typically combines careful tissue dissection with enzymatic digestion and mechanical dissociation to loosen cell-cell and cell-matrix connections while preserving cell viability and characteristic markers. Isolated epithelial cells support studies of barrier function, tissue development, regeneration, infection, and disease-related changes, including responses to drugs or environmental conditions. As a foundational technique in biology, it enables researchers to connect tissue architecture with cellular behavior and establish models that more closely represent epithelial physiology.

Analysis of vestibular hair cell function is complicated by their location deep within the hardest part of the skull, the petrous temporal bone. Most functional hair cell studies have used acutely isolated hair cells. Here we describe a semi-intact preparation of mouse vestibular epithelium for electrophysiological and two-photon microscopy studies.

This protocol details the separation of the mouse ocular lens epithelium and fiber cell bulk mass, followed by RNA isolation and qPCR analysis. This method allows for separating lens cell compartments for a more detailed analysis of the transcriptome and biological processes in epithelial cells vs. differentiated fiber cells.
