Amniotic Membrane Dissection

Amniotic membrane dissection is a biological technique used to isolate the amnion, the thin innermost layer of the fetal membranes, from surrounding placental tissues for examination or further processing. The procedure relies on careful separation along natural tissue planes, using controlled blunt or sharp dissection to preserve the membrane’s structure and attached cellular layers. Isolated amniotic membrane supports studies of extracellular matrix organization, epithelial and mesenchymal cell biology, development, and tissue repair. In research and clinical contexts, dissection also provides material for investigating barrier function, wound-healing properties, and potential regenerative applications.

This protocol describes the reprogramming of primary amniotic fluid and membrane mesenchymal stem cells into induced pluripotent stem cells using a non-integrating episomal approach in fully chemically defined conditions. Procedures of extraction, culture, reprogramming, and characterization of the resulting induced pluripotent stem cells by stringent methods are detailed.

Lumican Extraction from Amniotic Membrane and Determination of its Storage Temperature

Encephalopathy of prematurity encompasses the central nervous system abnormalities associated with injury from preterm birth. This report describes a clinically relevant rat model of in utero transient systemic hypoxia-ischemia and intra-amniotic lipopolysaccharide administration (LPS) that mimics chorioamnionitis, and the related impact of infectious stimuli and placental underperfusion on CNS development.
