Ex Vivo Heart Model

An ex vivo heart model is an isolated heart maintained outside the body to study cardiac structure, function, and responses under controlled laboratory conditions. Researchers typically preserve activity by perfusing the coronary vessels with an oxygenated, nutrient-containing solution, allowing heart rate, contractility, electrical activity, and vascular responses to be measured independently of whole-animal influences. In biology, this model supports investigation of cardiac physiology, ischemia-reperfusion injury, drug effects, and potential therapies while enabling precise control of experimental conditions. It provides a bridge between cellular studies and in vivo research, although the absence of systemic interactions limits how fully results represent a living organism.

Developmental studies in the mouse are hampered by the inaccessibility of the embryo during gestation. To promote the long-term culture of the embryonic heart at late stages of gestation, we developed a protocol in which the excised heart is cultured in a semi-solid, dilute Matrigel.

Most studies involving the Langendorff apparatus use small animal models due to the increased complexity of systems for larger mammals. We describe a Langendorff system for large animal models that allows for use across a range of species, including humans, and relatively easy data acquisition.

Antegrade Perfusion Model: An Ex Vivo Technique to Perfuse Isolated Mouse Heart

2025

This video describes an antegrade perfusion technique of an isolated mouse heart during which a perfusate is injected through the left ventricle, and eventually, it perfuses the entire myocardium. This method can be used to isolate cardiomyocytes from murine models and generate decellularized hearts for further downstream applications.

Here, we present a systematic approach for developing physiologically relevant, sensitive and specific in vivo assays for interpreting variation in human pathology. Transient genetic manipulation via microinjection of WT and mutant human mRNA and morpholino (MO) antisense oligonucleotides harness the tractability of the developing zebrafish embryo to rapidly assay pathogenic mutations, especially, but not exclusively, in the context of human developmental disorders.
