Exosome Purification

Exosome purification is the process of isolating exosomes, small extracellular vesicles released by cells, from complex samples such as biofluids or cell-culture media for biological analysis. Because these vesicles differ from contaminants in size, density, surface molecules, and interactions with specialized ligands, purification methods use principles such as differential centrifugation, size-exclusion chromatography, precipitation, or affinity capture. The resulting preparations support studies of intercellular communication, disease-associated biomarkers, and therapeutic delivery, while purification quality strongly affects downstream molecular profiling and functional assays. Comparing recovery, purity, scalability, and preservation helps researchers select an approach suited to each biological application.

Purification and microRNA Profiling of Exosomes Derived from Blood and Culture Media

The presence of stable microRNAs (miRNAs) in exosomes has generated immense interest as a novel mode of intercellular communication, for their potential utility as biomarkers and as a route for therapeutic intervention. Here we demonstrate exosome purification from blood and culture media followed by quantitative PCR to identify miRNAs being transported.

This paper demonstrates methods for the isolation, purification and detection of exosomes, as well as techniques for analysis of their molecular content. These methods are adaptable for exosome isolation from both cell culture media and biological fluids, and can beyond analysis of molecular content also be useful in functional studies.
