Matrigel Tube Formation

Matrigel tube formation is an in vitro assay that models an early stage of angiogenesis, the growth of new blood vessels, by measuring how endothelial cells organize into capillary-like networks. In the assay, endothelial cells are seeded onto a layer of Matrigel, a basement membrane extract rich in extracellular-matrix proteins and growth factors; cell adhesion, migration, alignment, and remodeling produce interconnected tubular structures within hours. Researchers quantify network length, branching, junctions, or total tube area to assess pro- or anti-angiogenic activity. The method supports studies of vascular biology, cancer, tissue repair, and drug screening, while providing a rapid complement to animal and three-dimensional models.

The tube formation assay is a fast, quantifiable method for measuring in vitro angiogenesis. Endothelial cells are combined with conditioned media and plated on basement membrane extract. Tube formation occurs within hours and newly formed tubules easily quantified.
