Fly Colony Housing

Fly colony housing is the controlled maintenance of flies in laboratory containers that support survival, reproduction, and consistent experimental conditions. In biology research, colonies are sustained by regulating temperature, humidity, light exposure, population density, and access to an appropriate nutrient medium while allowing adequate ventilation and preventing contamination or escape. Well-managed housing enables researchers to maintain stable stocks, collect individuals of defined age or genotype, and conduct studies of genetics, development, behavior, physiology, and disease. Standardized housing also improves reproducibility by reducing environmental variation and helps laboratories preserve valuable strains for ongoing experiments.

Source: Dodge, R., et al. Fast Colony Forming Unit Counting in 96-Well Plate Format Applied to the Drosophila Microbiome. J. Vis. Exp. (2023)This video demonstrates the quantification of colony-forming units (CFUs) of gut-associated bacteria in Drosophila flies using a serial dilution and plating approach. Homogenized fly samples containing gut bacteria are serially diluted, plated on agar, and incubated to allow colony growth. Images of the plates are then analyzed using software to align,...

Counting Colony Forming Units (CFUs): Quantifying Bacteria from Fly Bodies

2023

Drosophila microbiota impacts the animal's development, immunity and physiology. This video describes a method to quantify bacteria from flies by determining the colony forming units from whole body preparations. The featured protocol demonstrates the technique with flies, reared under gnotobiotic conditions using four bacterial species.

This protocol describes a microinjection methodology that we have standardized and used for several years to deliver specific quantities of nucleic acids directly to the hemolymph of mosquitoes and house flies. This protocol results in minimal injection mortality and allows dose correlated measurements of fecundity.

Here, we present a protocol to produce house fly carboxylesterase proteins in vitro with a baculovirus mediated insect cell expression system and later functionally characterize their roles in metabolizing permethrin, thereby, conferring pyrethroid resistance by conducting cell-based MTT assay and in vitro metabolic studies.

Drosophila Courtship Conditioning: A Method to Test Learning and Memory In Flies

2023

This video describes a classical conditioning assay that tests learning and memory in Drosophila, called courtship conditioning. The assay is based on the reduction of male courtship after experiencing a rejection by a non-receptive premated female. The example protocol shows a setup for the procedure that can be used to assess short- and long-term memory.
