Glucose Phosphorylation

Glucose phosphorylation is the addition of a phosphate group to glucose, a fundamental reaction that regulates how cells retain and use this important energy source. In the first step of glycolysis, enzymes such as hexokinase or glucokinase transfer a phosphate group from ATP to glucose, producing glucose-6-phosphate; the charged product remains inside the cell and can enter metabolic pathways. This reaction supports energy production, glycogen synthesis, and the pentose phosphate pathway. Studying glucose phosphorylation helps explain cellular metabolism, tissue-specific fuel use, and disorders in which altered glucose handling contributes to diabetes and other metabolic diseases.

The addition or removal of phosphate groups from proteins is the most common chemical modification that regulates cellular processes. These modifications can affect the structure, activity, stability, and localization of proteins within cells as well as their interactions with other proteins. During phosphorylation, protein kinases transfer the terminal phosphate group of ATP to specific amino acid side chains of substrate proteins. Serine, threonine, and tyrosine are the most commonly...

In this video, we demonstrate an ex vivo technique to measure extracellular glucose analog depletion in harvested mouse organs. It involves incubating the mouse tissue in a solution containing fluorescently-labeled deoxyglucose, which is taken up by the cells. The glucose analog uptake can be indirectly determined by measuring the fluorescence of the incubated medium.
