Intracellular Receptors

Intracellular receptors are proteins located within cells that detect small, membrane-permeable signaling molecules and regulate gene activity, making them central to cellular communication and physiological control. Lipophilic ligands such as steroid hormones cross the plasma membrane, bind receptors in the cytoplasm or nucleus, and trigger receptor activation, DNA binding, and recognition of specific hormone response elements that alter transcription. In biology, intracellular receptors help explain endocrine signaling and the regulation of development, metabolism, and homeostasis. Their selective interactions with ligands also make them important targets for studying disease mechanisms and developing therapeutic drugs.

Localization of Plasma Membrane and Intracellular Neuronal Nicotinic Acetylcholine Receptors Using Quantitative Imaging in Mammalian Cells

This protocol describes expression and quantification of neuronal nicotinic acetylcholine receptors subunits using the mammalian Neuro-2a cell line. pH-sensitive fluorescent tags and confocal microscopy enable precise evaluation of receptor localization at the plasma membrane versus those expressed in pH-neutral intracellular compartments, facilitating the study of nAChR trafficking and pharmacological modulation.
