Marine Microbial Dna

Marine microbial DNA is the genetic material found in microorganisms inhabiting oceans, coastal waters, sediments, and other marine environments, providing a molecular record of microbial diversity and function. Its nucleotide sequences encode inherited traits and can be isolated from cells or environmental samples, then amplified, sequenced, and analyzed to identify organisms and genes. These methods support marine microbial ecology by revealing community composition, metabolic capabilities, and responses to changing conditions. Studying marine microbial DNA also helps researchers investigate nutrient cycling, climate-related processes, harmful algal events, and potential sources of enzymes or other biologically valuable molecules.

We present a method for generating cDNA from environmental mRNA. In general, total RNA is first collected from the environment, rRNA is selectively removed, mRNA is selectively amplified, and cDNA synthesized from the enriched mRNA pool is sequenced. Recovered sequences can be annotated using standard bioinformatics techniques to identify the expressed genes.

Extraction of High Molecular Weight DNA from Microbial Mats

We provide an improved protocol for extracting high molecular weight DNA from hypersaline microbial mats. Microbial cells are separated from the mat matrix prior to DNA extraction and purification. This enhances the concentrations, quality, and size of the DNA. The protocol may be used for other refractory samples.
