Confocal Scanning Microscopy

Confocal scanning microscopy is an optical imaging technique that produces high-resolution, depth-resolved images by collecting light from a focused point within a specimen while rejecting out-of-focus signals. A laser scans the sample point by point, and a spatial pinhole blocks emitted or reflected light that does not originate from the focal plane; computer software then reconstructs the detected signal into optical sections and three-dimensional images. In biology, confocal scanning microscopy is used to examine fluorescently labeled cells, tissues, and subcellular structures with improved contrast and spatial resolution. It supports studies of cell organization, protein localization, tissue architecture, and dynamic biological processes.

Video-rate Scanning Confocal Microscopy and Microendoscopy

The complete construction of a custom, real-time confocal scanning imaging system is described. This system, which can be readily used for video-rate microscopy and microendoscopy, allows for an array of imaging geometries and applications not accessible using standard commercial confocal systems, at a fraction of the cost.

We present a protocol for structural and compositional analysis of natural oral biofilm from orthodontic appliances with in situ hybridization (FISH) and confocal laser scanning microscopy (CLSM). Oral biofilm samples were collected from palatal expanders, scraping acrylic-resin flakes off their surface and referring them for molecular processing.
