Plant Tissue Imaging

Plant tissue imaging is the use of microscopy and related optical methods to visualize the organization, structure, and activity of cells within plant tissues. By transmitting or detecting light from intact or prepared samples, often with stains, fluorescent markers, or optical sectioning, researchers generate two- and three-dimensional images that reveal features such as cell boundaries, vascular tissues, organelles, and molecular signals. These observations support studies of plant development, growth, transport, stress responses, and disease, while quantitative image analysis can measure spatial patterns and changes over time. Plant tissue imaging therefore connects cellular mechanisms with whole-organ function in biology.

Robotics and Dynamic Image Analysis for Studies of Gene Expression in Plant Tissues

We report a method for introduction, tracking and quantitative analysis of GFP expression in plant cells. This method utilizes a custom-designed robotics system for semi-continuous image collection from large numbers of samples, over time. We also demonstrate the use of ImageJ and ImageReady for analysis of image series.

This video demonstrates an immunostaining technique to localize arabinogalactan proteins (AGPs) and pectins in plant tissues. Fixed and resin-embedded plant tissue sections are stained with AGP- and pectin-specific primary and fluorescent secondary antibodies and a cellulose-binding fluorescent dye to reveal their tissue-specific colocalization in the cell wall.
