Toxin Induced Damage

Toxin-induced damage is the disruption of cellular structure or function caused by toxic substances, ranging from microbial products and venoms to environmental chemicals and drugs. Toxins can act by binding specific receptors or enzymes, generating reactive oxygen species, damaging membranes or DNA, and overwhelming protective systems such as antioxidant and repair pathways; the resulting injury may trigger apoptosis, inflammation, or tissue failure. In biology, studying these mechanisms helps researchers explain disease, assess environmental and pharmaceutical hazards, identify biomarkers of exposure, and develop antidotes or protective treatments. Experimental models connect molecular lesions with effects across cells, organs, and organisms.

Methods for purifying the cholesterol binding toxin streptolysin O from recombinant E. coli and visualization of toxin binding to live eukaryotic cells are described. Localized delivery of toxin induces rapid and complex changes in targeted cells revealing novel aspects of toxin biology.

Inducible Expression of Histidine-Tagged Ricin Toxin A Chain in E. coli

Source: Becker, B., et al. A Simple Fluorescence-based Reporter Assay to Identify Cellular Components Required for Ricin Toxin A Chain (RTA) Trafficking in Yeast. J. Vis. Exp. (2017)This video demonstrates the inducible expression of His-tagged ricin toxin A chain (RTA) in Escherichia coli. It outlines the steps involved in the selective culturing of bacteria transformed with a recombinant expression plasmid, induction of His-tagged RTA expression, and preparation of the bacteria for further...

Assessing DNA Damage Foci: To Identify Radiation-Induced DNA Damage Foci in Cancer Cell Line Using Immunofluorescence Staining

2023

This video describes a protocol to detect radiation-induced DNA repair foci in the colon cancer cell lines using immunofluorescence staining. DNA repair proteins get activated at the DNA damage site; therefore, we can detect them using specific antibodies and visualize them under a fluorescence microscope.

Hematoxylin and Eosin Staining to Assess Colon Damage in DSS-Induced Colitis

This video demonstrates the hematoxylin and eosin (H&E) staining method to evaluate colon damage in a murine model of DSS-induced colitis — an inflammatory bowel disease. Upon isolating the colon from a DSS-treated mouse and obtaining sections of the tissue, H&E staining of the sections helps identify the inflammation-mediated tissue damage.

Imaging Brain Damage Induced by Cerebral Hypoxia-Ischemia in a Mouse Model

Source: Ouyang, Y., et al. Simultaneous PET/MRI Imaging During Mouse Cerebral Hypoxia-ischemia. J. Vis. Exp. (2015)This video demonstrates a technique to assess cerebral hypoxia injury via simultaneous positron-emission tomography (PET) and magnetic resonance imaging (MRI). An anesthetized mouse undergoes common carotid artery (CCA) ligation, reducing blood flow to one brain hemisphere and predisposing it to hypoxia-induced injury. PET and MRI imaging evaluate glucose metabolism and structural...
