Spinoculation

Spinoculation is a centrifugation-assisted method that improves the delivery of viral vectors into cultured cells, making it useful for experimental gene transfer. During spinoculation, cells and vector particles are incubated together while centrifugation brings the particles into closer contact with the cell surface, which can increase vector attachment and subsequent entry. In cancer research, this approach supports the generation of genetically modified tumor cell models for studying oncogenes, tumor suppressor pathways, drug responses, and immune-cell recognition. By improving transduction efficiency or reducing the amount of vector needed, spinoculation can make gene delivery more consistent across challenging cell types, although outcomes depend on cell state, vector design, and centrifugation conditions.
