Gallium-68 Tracer

A Gallium-68 tracer is a radiolabeled chemical compound that uses the positron-emitting radionuclide gallium-68 to make molecular processes visible with positron emission tomography (PET). In radiolabeling, gallium-68 coordinates with a chelator, such as a DOTA-based ligand, which is attached to a targeting molecule and stabilizes the metal complex under physiological conditions. After administration, the targeting component directs the tracer toward specific receptors or tissues, while positron decay produces signals reconstructed into images. Gallium-68 tracers support noninvasive studies of tumor biology, receptor expression, and disease distribution, linking coordination chemistry with diagnostic imaging and precision medicine.

This study developed a noninvasive and real-time method to evaluate the distribution of programmed death-ligand 1 in the whole body, based on positron emission tomographic imaging of [68Ga] D-dodecapeptide antagonist. This technique has advantages over conventional immunohistochemistry and improves the efficiency of identifying appropriate patients who will benefit from immune checkpoint blockade therapy.
