Fibrin Gel Scaffold

A fibrin gel scaffold is a three-dimensional, biologically derived matrix that supports cell organization, survival, and tissue formation, making it valuable for studying development and regeneration. It forms when thrombin cleaves fibrinogen, allowing fibrin molecules to polymerize into an interconnected network whose density, stiffness, and degradation can be adjusted during preparation. In developmental biology, this scaffold provides cells with adhesion sites and a tissue-like environment that can influence migration, differentiation, and morphogenesis. Researchers use fibrin gels to culture embryonic or progenitor cells, model extracellular-matrix interactions, and investigate how physical and biochemical signals shape developing tissues.

Clot Retraction and Fibrinolysis

After a fibrin clot is formed, the next step is clot retraction, a vital process facilitated by platelet contractile proteins, such as actin and myosin. These proteins pull the fibrin strands closer together and condense the clot. This action reduces the size of the clot, creating a smaller, denser structure that effectively seals off the damaged vessel. Clot retraction consolidates the clot and helps with wound healing by bringing the edges of the damaged blood vessel closer together.

Here, we present a protocol to fabricate electrospun nanofiber scaffolds with gradated organization of fibers and explore their applications in regulating cell morphology/orientation. Gradients with regard to physical and chemical properties of the nanofiber scaffolds offer a wide variety of applications in the biomedical field.
